HISTORATHOLOGICAL AND HISTOCHEMICAL STUDIES ON THE GINGIVAE IN PERIODONTITIS

Abstract
Histopathological and histochemical studies were performed to observe the local tissue

responses, the distributions of mucopolysaccharide, RNA and DNA by inflammatory reaction, the relationship between inflammation and keratinization.
In these studies, thirty specimens taken from the gingivae in periodontitis which had collected from .April, 1971 to. July, 1971 were used.
The biopsies were prepared according to the Harris¢¥ Hematoxylin-Eosin stain, the w Mallory¢¥s Aniline Blue stain, the Hotchkiss¢¥, Periodic Acid-Schiff stain, the Lillie¢¥s Me-
thyl Green-Pyronin stain, the Feulgen¢¥s DNA stain.
The results were as follows.

1. On the whole, the slightly, inflammed cases revealed the keratinized layer and/or the
t parakeratinized layer, but- the severe cases did not reveal the keratinization. With j
the Mallory¢¥s Aniline Blue Stain, the keratinized layer was deeply stained showing the
regular lines and/or irregular masses but the parakeratinized layer was stained - faintly and irregularly.
2. With severity of inflammation, hydropic degeneration of epithelium was increased ¢¥ . and collagenous fibers in lamina propria appeared much less:
3. With the Hotchtiss¢¥ Periodic Acid-Schiff stain the keratinized layer was reacted it
faintly, the parakeratinized layer, granular. layer and prickle cell layer intensely,


basal cell Layer¢¥ faintly or negatively, basement membrane `intensely, fibers¢¥ around
blood vessels and collagenous fibers in lamina propria intensely. In the severe cases, the degree of PAS reaction was weakened as compared with the slight cases.
4. With the Lillie¢¥s Methyl Green-Pyronin Stain, basal cell layer was deeply stained, showing a gradual diminution of staining intensity to the outer surface of epithelium and the keratinized layer was not stained with Methyl Green, but deeply stained with Pyronin. In the severe cases, the degree of the stain was diminished as compared with the slight cases.
5. With Feulgen¢¥s DNA stain, the results were similar with those of Methyl Green, but pyknosed nuclei in parakeratinized layer was stained deeply. The degree of the stain in severe cases revealed weakened tendency as compared with the slight cases.
